Fibroblasts and keratinocytes play a major role in the process of wound healing. Hence, wound healing properties of compounds are studied by inflicting wound over the Fibroblast (Swiss 3T3)/Keratinocyte (HaCaT) monolayer and monitoring the rate of recovery.
The 24 hour monolayer in a 24 well plate (2 X 10(4) seeding density of the cells) was scratched using a 1ml pipette tip. The medium (DMEM) is then replaced with fresh medium containing varying concentrations of the sample. The cell proliferation was observed at regular intervals of 24hrs. The results are expressed as percentage recovery at a given dosage with pictorial representations
Result: Varying concentrations of 12.5, 25, 50 and 100µg/ml had been analyzed for Wound healing activity. At concentrations of 12.5, 25, 50µg/ml, a significant and constant enhancement of 25% in wound healing activity has been observed.

The epidemic of cholera on the Pacific Ocean atoll of Tarawa, Gilbert Islands renewed interest in the use of coconut water as a rehydration fluid. Fifty-one samples of coconut water from Tarawa were analysed for a variety of constituents to assess its potential usefulness in the oral and parenteral rehydration of patients with cholera and other severe forms of gastroenteritis. Compared to oral rehydration fluids known to be effective in cholera, coconut water was found to have adequate potassium and glucose content, however was relatively deficient in sodium, chloride and bicarbonate. The addition of table salt to the coconut water is suggested to compensate for the sodium and chloride deficiency. In areas of the world where coconuts are plentiful, the advantages of sterility, availability and acceptability make coconut water theoretically feasible for the oral rehydration of patients with severe gastroenteritis when conventional fluids are unavailable

The effect of coconut water (CW) on biomass and docosahexaenoic acid (DHA, C22:6 n3) formation by Schizochytrium mangrovei Sk-02 was studied in a yeast extract-diluted sea water medium. Optimal CW-level was ca. 33% (v/v), resulting in a biomass level of 28 g/l with a DHA-content of 20% (w/w) or 6 g DHA/l, almost 50% higher than in non-supplemented cultures at the same initial sugar level. Study on the growth-promoting effects of coconut water suggested that it could be (partially) mimicked by addition of trace elements; the fatty acids present in CW did not appear to be incorporated or effect fatty acid formation by the organism. CW-addition was also effective in media with other nitrogen sources such as casitone, peptone and tryptone. Its inclusion (at 50% v/v) increased biomass levels two-to-three-fold with concomitant increases in the DHA-level.
