Alpinia galanga

EnXtra™ from Alpinia galanga is a DNA authenticated, stim-free energy ingredient that provides fast, effective and safe energy with no crash. In a 53-person, double-blind, double-dummy human clinical trial, It was shown to improve mental alertness & acuity for up to 5 hours without increasing heart rate or blood pressure. It can be used as a replacement for caffeine or used with caffeine to prevent crash and prolong benefits. EnXtra is easy to formulate with, water soluble and has a pleasant taste profile.

EnXtra™ is a DNA-authenticated galangal extract, Alpinia galanga, a genus of perennial herbs, with rhizomatous rootstocks and tall, leafy stems found in warmer climates in Asia.
The roots of commonly grown species of Alpinia appear similar to untrained eyes, and any adulteration and substitution would go unnoticed. EnXtra’s authentication process is a long journey involving collecting samples from farms in southern parts of India and testing them with modern scientific methods. When the fingerprint of biomass, in comparison with authentic control sample has more than 99% specificity i.e. confirming DNA authenticity, only then the rhizomes are qualified to make EnXtra.

To better understand how EnXtra provides 5hrs of Mental Alertness & Sustain attention, without the crash, a molecular docking study of a database of molecules of EnXtra was performed against a panel of target proteins responsible for enhancing attention network components. The binding efficacies of these compounds were computed using a parameter called “Glide score”. Glide Score negative values representing tighter binding. A Glide score < minus 6 is considered good binding capacity of the ligand (molecule) to target proteins.

During a randomized, double-dummy, double blind, placebo-controlled crossover study, which was primarily designed to elucidate the Investigational Product (IPs’) (EnXtra and caffeine) effect on various psychoactive measures in habitual caffeine consumers, it was observed that EnXtra showed improvement in the alertness scores until 5 hours, whereas caffeine which is well-known for its effect on the alerting network, showed increase until 1 hour, followed by a reduction—probably due to a caffeine crash—at 3 hours. The Java version of the ANT 1.3.0 was used for this study, as it provides a behavioral measure of efficiency of the different components of the attention network separately within a single task. In addition, the probability of a combined effect to reduce the caffeine crash was explored by the co-administration of EnXtra and caffeine. In this composite group, alertness scores increased significantly at 1 hour followed by a reduction, indicating a caffeine crash, but this was less than that observed in the caffeine only group. Hence, it can be suggested that EnXtra is able to impede the caffeine crash, as evident from 3-hour and 5-hour alertness scores.
A new 90-day clinical study to further prove long term efficacy and safety of EnXtra is also under process.

Mechanism of Action : An in-silico study (molecular docking study) was conducted with the objective of targeting proteins known to have significant role in neurostimulation mechanisms such as dopamine reuptake and acetylcholine esterase inhibition.
This study demonstrated that active compounds of EnXtra® increase dopamine levels by blocking the uptake.
This increases dopamine levels which enhances the visuospatial performance and mental clarity leading to improved mental alertness.
Also, the study revealed EnXtra’s potential as a nootropic agent by its ability to inhibit Acetylcholine-esterase (AchE) target protein.
Hence it can be concluded that bioactive molecules from EnXtra® decrease dopamine uptake and inhibit acetylcholine esterase activity thereby producing the alertness enhancing and focus effects.
